The annealing temperature (Ta) is the critical step in PCR where primers bind to the target DNA. This temperature must be balanced: high enough to ensure primers bind only to the perfect match (specificity), but low enough for efficiency. This calculator uses the Rychlik formula, which considers both the primer and the product stability.
